实验感染肝片吸虫对大鼠肝脏药物代谢功能的影响 被引量:3

Effects of experimental fascioliasis on the drug-metabolizing function in rat liver

摘要 将 8 5只体重 2 5 0~ 35 0 g的健康SD大鼠 (雌雄各半 )随机分成感染组 (n =5 5 )和对照组 (n =30 )。感染组大鼠每只一次经口感染 2 5个肝片吸虫囊蚴 ,每周采血测定血浆天冬氨酸转氨酶 (AST)、丙氨酸转氨酶 (ALT)及碱性磷酸酶 (ALP)活性。于感染后第 3,6 ,9,11和 13周分别宰杀 12只 (感染组 8只 ,对照组 4只 )大鼠 ,测定其肝微粒体蛋白、细胞色素P 4 5 0、细胞色素b5的浓度和肝脏药物代谢酶活性。结果表明 ,大鼠感染肝片吸虫后 ,血浆AST、ALT、ALP活性分别在感染后 3~ 7周、 3~ 5周、 5~ 13周极显著上升 (P <0 0 1) ;肝体比上升 ;CytP 4 5 0浓度、肝脏药物代谢酶活性极显著下降 (P <0 0 1)。提示肝片吸虫感染对大鼠肝脏药物代谢功能有明显的损伤作用。 Eighty-five SD rats weighting 250350 g were used and divided randomly into infection group(n=55)and control group(n=30).Each infected rat received 25 metacercariae of Fasciola hepatica by gastric tube.AST,ALT and ALP activities in plasma were determined each week.12 rats(8 infected and 4 uninfected animals)were killed at 3,6,9,11,13 weeks post-infection,respectively.The levels of liver microsomal protein,cytochrome P-450(Cyt P-450)and b 5(Cyt b 5 ),liver drug-metabolizing enzymes were determined,respectively.The results showed that the levels of AST,ALT and ALP and liver/body weight ratio were increased significantly(P<0.01)at 37,35,513 weeks post-infection of Fasciola hepatica,the levels of cytochrome P-450,aminopyrine N-demethylase(AND),aniline 4-hydroxylase(AH),UDP-glucuronyltransferase(UGT)and glutathione S-transferase(GST)reduced(P<0.05),the drug-metabolizing function in rat liver was demaged because of F.hepatica infection.
