摘要
目的 探讨皮下脂肪细胞摄取氧化低密度脂蛋白 (Ox -LDL)的可能机制和影响因素。方法 用高胆固醇饮食法建立动脉粥样硬化模型 ,将喂食高胆固醇兔随机给予 1 5mg·kg- 1·d- 1的阿托伐他汀 (n =5)或淀粉 (n =5) ,2周后取腹股沟处皮下脂肪组织进行脂肪细胞培养。测定细胞对12 5 I OxLDL的摄取情况。并检测过氧化物酶增殖型激活受体 (PPAR)γ和清道夫受体 (CD3 6)mRNA的表达情况。结果 正常组脂肪细胞对12 5 I OxLDL的特异性摄取量明显高于阿托伐他汀组和淀粉组 [(82 9± 3 0 )比 (682± 52 )和 (3 62± 2 6)ng/mg·细胞蛋白 ,P <0 0 5]。脂肪细胞对12 5 I OxLDL的特异性摄取量与PPARγmRNA(r =0 660 ,P =0 0 14 )和CD3 6mRNA (r =0 80 2 ,P <0 0 1)的表达以及血浆低密度脂蛋白胆固醇水平 (r = 0 855,P <0 0 1)相关。结论 兔皮下脂肪细胞具有摄取Ox LDL能力。高胆固醇血症时 ,皮下脂肪细胞对Ox LDL的摄取能力减弱 ,阿托伐他汀可能通过降低血胆固醇水平和调节PPARγ、CD3 6的表达来改善脂肪细胞对Ox
Objective Peroxisome prolif erator-activated receptors (PPAR) γ and CD36 receptor, a plausible pathway for oxidized low-density lipoprotein (Ox-LDL) uptake in monocytes, highly express in adipocytes In this study, we sought to test whether subcutaneous adipocytes could uptake Ox-LDL and tried to explore possible mechanisms for the Ox-LDL uptake in adipocytes from hypercholesterolemic rabbits Methods Fourteen male New Zealand rabbits were randomly divided into normal diet group (n=4), 1 2% high-cholesterol diet group (n=10) After 8 weeks, hypercholesterolemic rabbits were switched to receive either atorvastatin 1 5 mg·kg -1 ·d -1 (n=5) or starch (n=5) Two weeks later, subcutaneous adipose was collected from groin of all rabbits under deep anesthesia to culture adipocytes After increasing concentrations of 125 I-OxLDL incubations, the 125 I-OxLDL uptakes in adipocytes were measured by gamma counter and normalized to the protein concentration of each sample Semi quantity reverse transcription polymerase chain reaction was used to determine PPARγ and CD36 mRNA expressions in adipocytes Results The specific uptaking value of 125 I-OxLDL in adipocytes was significantly higher in normal diet group than those in atorvastatin or starch treated groups (829±30 vs 682±52 and 362±26 ng/ mg cell protein, P<0 05) The specific uptakings of 125 I-OxLDL in adipocytes were intimately correlated with mRNA expressions of PPARγ (r =0 660, P=0 014) and CD36 (r=0 802, P<0 01) as well as plasma LDL-C level (r=0 802,P<0 01) Conclusion The results indicate that rabbit subcutaneous adipocytes can uptake Ox-LDL Hypercholesterolemia decreases the Ox-LDL uptake in adipocytes, and atorvastatin could improve the Ox-LDL uptake through reducing plasma cholesterol level and enhancing mRNA expressions of PPARγ and CD36 receptor in adipocytes
