摘要
【目的】探讨青蒿琥酯对人肺腺癌H1975细胞株增殖、迁移及侵袭的影响及其可能机制。【方法】不同浓度青蒿琥酯干预肺腺癌H1975细胞24 h后,细胞计数试剂盒-8(cell counting Kit-8,CCK-8)实验检测细胞的增殖能力,并计算IC50值。设置不加药组为空白对照组,根据IC50值选取20、40、80μg/ml 3个浓度干预H1975细胞作为实验组,划痕实验检测细胞迁移能力,Transwell实验检测细胞侵袭能力,RT-qPCR和Western blotting检测趋化因子受体4(chemokine receptor 4,CXCR4)的mRNA和蛋白质表达。【结果】CCK-8实验结果显示青蒿琥酯对H1975细胞株增殖有抑制作用,且呈浓度依赖性,IC50值为95.35μg/ml。划痕实验、Transwell实验提示实验组各浓度梯度的青蒿琥酯处理H1975细胞24 h后均能抑制细胞的迁移、侵袭能力。各实验组划痕愈合百分比(分别为37.67%±4.04%、24.00%±1.00%和16.33%±1.53%)与空白对照组(61.33%±5.13%)相比明显降低(P<0.01)。穿膜细胞数(分别为78.33±2.08、45.00±8.18和16.33±5.15),与对照组(78.33±2.08)相比明显减少(P<0.01)。RT-qPCR实验提示各实验组细胞CXCR4 mRNA相对表达(0.704±0.080、0.483±0.008和0.404±0.056)与空白对照组相比(1.073±0.083),明显减少(P<0.05)。Western blotting实验提示各实验组CXCR4的蛋白的相对表达量(0.722±0.073、0.585±0.029和0.420±0.036)和空白对照组相比(0.214±0.021)明显下降(P<0.05)。【结论】青蒿琥酯能抑制肺腺癌H1975细胞增殖、迁移及侵袭,其机制可能与下调CXCR4的表达有关。
【Objective】To explore the effects and its potential mechanism of artesunate on the proliferation,migration and invasion of lung adenocarcinoma cell line H1975.【Methods】After lung adenocarcinoma H1975 cells were treated with different concentrations of artesunate for 24 h,CCK-8(Cell Counting Kit-8)assay was used to detect the proliferation of cells,and the IC50 value was calculated.The non-medicated group was set as a blank control group,and the H1975 cells were treated with three different concentrations of 20,40,80μg/ml according to the IC50 values as the experimental groups.The scratching test was used to detect the cell migration ability,and the transwell assay was used to detect the cell invasion ability,RT-qPCR and Western blot were used to detect the mRNA and protein expressions of Chemokine receptor 4(CXCR4).【Results】The results of CCK-8 assay showed that artesunate inhibited the growth of H1975 cells in a dose-dependent manner and IC50 value was 95.35μg/ml.The results of scratching test and transwell assay indicated that artesunate in each concentration gradient could inhibit the migration and invasion ability of H1975 after 24 h.The wound healing percentages of experimental groups(37.67%±4.04%,24.00%±1.00%and 16.33%±1.53%,respectively)were significantly lower than that in the blank control group(61.33%±5.13%,P<0.01).The numbers of membrane-penetrating cells in experimental groups(78.33±2.08,45.00±8.18 and 16.33±5.15,respectively)were significantly reduced compared with the blank control group(78.33±2.08,P<0.01).The relative expressions of CXCR4 mRNA in experimental groups(0.704±0.080,0.483±0.008 and 0.404±0.056)were significantly down-regulated compared with the blank control group(1.073±0.083,P<0.05).The CXCR4 protein expression levels in experimental groups(0.722±0.073,0.585±0.029 and0.420±0.036)were significantly down-regulated compared with the blank control group(0.214±0.021,P<0.05).【Conclusion】Artesunate can restrain the proliferation,migration and invasion of H197
