摘要
为保证保健食品的质量 ,建立了测定食品中金雀异黄素的HPLC方法。试样以乙醚脱脂 ,甲醇 +水 (80 + 2 0 ,体积分数 )超声提取 30min ,定容后过 0 4 5 μm滤膜 ,上 2 5 0 0mm× 4 6mm、粒径10 μm的C18柱 ,甲醇 +醋酸铵 (0 0 5 0mol L ,pH 4 6 )作流动相 ,二极管阵列检测器测定。染料木苷、黄豆苷、黄豆苷元对测定不产生干扰。方法回收率 85 %~ 110 % ,相对标准偏差 2 %~ 8% ,最低检出限为 0 10 μg。本方法适用于保健食品中金雀异黄素的测定。
A rapid method using a reverse-phase high performance liquid chromatography for the determination of genistein in foods was presented.After the sample was sonicated for 30 minutes and passed through a 0.45 μm filter,it was then separated on a C_ 18 reversed-phase column (25 cm×4.6 mm i.d.;10 μm particles) and methanol—0.050 mol/L ammonium acetate (46%+54%,by volume),pH 4.6 was used as the mobile phase at a velocity of 1.2 mL/min.The eluate was detected by a photo diode-array detector at 210 nm~400 nm.The separation of genistein,genistin,daidzein and daidzin was achieved successfully in about 20 minutes.The recovery of genistein was 85%~110% and the variation coefficient was 2%~8%.This method was suitable for the determination of genistein in foods.
