海马 CA_3区神经元的 RNA、衰老色素、核仁体积和细胞数的年龄变化 被引量:4

摘要 用形态计量学,组织化学和荧光显微定量的方法研究4个年龄组(1,3,17,30个月)Wistar 大鼠的海马 CA_3区锥体细胞的 RNA、衰老色素(Lp)、核仁体积和细胞数(细胞数/250μm)的增龄变化。结果 CA_3神经元的 RNA、核仁体积和细胞数随增龄而进行性减少,以17~30个月减少最明显,差异有高度显著意义(P<0.01)。细胞内 Lp 累积随年龄增加而不断增多,各相邻年龄组之间差异均有高度显著性(P<0.01)。对40例大鼠的 RNA 含量和神经元数分别与 Lp 含量进行直线相关和回归分析,结果是随着 Lp 含量的增加 RNA 含量不断减少:r=-0.86(P<0.01),Y=221.67-1.16X;神经元数随 Lp 增加而减少:r=-0.98(P<0.01),Y=54.98-0.20X。本文还对海马神经元的 RNA 减少,Lp 累积和细胞丢失的意义及其与老年性记忆减退的关系进行了讨论。 The methods of morphometry,histologic fluorescence quantification and quantita- tive histochemistry,RNA content,lipofuscin (Lp) content,nucleolar volume and neuron number (cells/250μm) were applied in the study of neurons in hippocampal CA_3 area in 4 age groups of Wistar rats (1,3,17,30 months). RNA content,nucleolar volume and neuron number decrease with advancing age.The difference between adult (17 months) and aged (30 months) rats were significant (P<0.01) for all of the 3 parameters mentioned aboved.However,Lp content increase with advancing age,which were significantly different among the four groups (P<0.01).Analysis liner regression and simple correlation shows that RNA content decrease with Lp content increasing:r=-0.86 (P<0.01),Y=221.67- 1.16X,and that neuron number reduce with Lp content increasing:r=-0.98 (P< 0.01),Y=54.98-0.20X.In this paper we disscused the significance of the changes of RNA and Lp,and neuron loss,as well as their relations to senile memory deficit.
