摘要
将自体坐骨神经移植到成体金黄地鼠视皮层17/18a 交界附近,或靠近枕部大脑的中缝处。手术42~131天后,在移植的外周神经断端,给以 HRP(浓度50%,溶于生理盐水中),观察HRP 逆行标记的再生视皮层神经元。发现外周神经移植法可诱导成体金黄地鼠视皮层神经元再生;各种大小不等、形状各异的神经元,都可再生出纤维到移植的外周神经中,长达10mm;再生神经元分布于靠近移植神经的皮层Ⅱ~Ⅵ层中;某些再生的神经元胞体,较正常细胞明显增大,这与金黄地鼠视网膜神经节细胞轴突再生的情形类似,但金黄地鼠视皮层神经元再生能力远不如视网膜神经节细胞。本实验用荧光双标记法,证实皮层视区胼胝体神经元是可以再生的。用“预先溃变”和“额外损伤”法,未能导致视皮层再生神经元数目的增加。本实验结果还表明,外周神经移植法可诱导金黄地鼠视皮层神经元再生,局部环境因素是影响哺乳动物中枢神经元再生能力的重要原因之一。
A segment of autologous sciatic nerve was transplanted into the 17/18a border
oregion or near the midline of the occipital brain in the adult golden hamsters.After
42-131 days survival time,HRP (50% in saline) was administered to the cut end
of the graft.It was found that the neurons of different sizes and shapes in visual
cortex can be induced to regenerate their fibers into the peripheral nerve graft for
up to 10mm;Some of the regenerated cells became larger than that of the normal cells.
in the visual cortex,which is similar to that in the retina,but the ability of regenera-
tion of the cortical neurons is much lower than that of the retinal ganglion cells.
Fluorescent dye double-labelling method was used and it was demonstrated that
the callosal neurons in visual cortex can regenerate their fibers into peripheral nerve
graft,the regenerative fibers originated from axotomized neurons rather than from
collateral sprouting of undamaged neurons.Predegeneration treatment for the sciatic
nerve did not enhance the regeneration of neurons in the visual cortex.Extradamage
made near the graft insertion does not seem to increase the quantity of regeneration
cells in the visual cortex.
The results of the experiment suggest that peripheral nerve graft can induce
regeneration of neurons in visual cortex and the local environment is one of the
important factors affecting the ability of regeneration of mammalian CNS.
