摘要 目的 探讨弹性蛋白酶在球囊损伤后动脉中的表达。方法  2F球囊导管损伤Wistar大鼠主动脉。用大鼠胰腺丝氨酸弹性蛋白酶Ⅱ的RNA探针进行原位杂交和Northern印迹杂交、抗弹性蛋白酶抗体进行免疫组化、胰腺弹性蛋白酶底物测定其活性 ;电镜观察迁徙细胞与其周围弹性纤维的连接处的数量。结果 球囊损伤后 ,迁徙细胞弹性蛋白酶mRNA、蛋白表达及其活性均有明显增加 ,细胞周围弹性纤维的连接处数量显著减少。结论 弹性蛋白酶是降解细胞外基质和动脉血管内弹性板、促使平滑肌细胞向内膜迁徙的重要原因。 Objective: To investigate the expression of the elastase in the artery after balloon injury. Methods:The aortas of male Wistar rats were injured by 2F balloon catheter, in situ hybridization and Northern blotting were performed by DNA probe of rat pancreas serine elastase Ⅱ. Immunostaining was made with antibody against elastase.The pancreatic elastase activity was assayed with given substrate.The contact number of the migrating cells with elastic fiber was observed by transmission electron microscopy.Results: The expression of elastase mRNA, its protein and activity were obviously increased after balloon injury.The contact number of elastic fibers surrounding normal medial smooth muscle cells obviously decreased. Conclusions: Elastase is an important factor that degrades extracellualr matrix, arterial internal elastic lamella and promotes migration of smooth muscle cells into the intima.
