摘要
目的 :建立黄芩苷HPLC测定方法 ,并通过比较研究小柴胡汤 (SST)不同提取部位及配伍对黄芩苷的影响 ,为制定黄芩苷质量标准提供参考依据。方法 :用水提醇沉、氯仿、正己烷萃取等方法提取小柴胡汤中的有效成分 ,并按配伍原则分解水煎提取小柴胡汤 ,得到不同分解提取剂。应用高效液相色谱法 (HPLC)测定其中黄芩苷的含量变化。结果 :提取剂中 ,6 0 %乙醇提取剂中黄芩苷的苷药比最高 (3 4 5 % ) ;正己烷、氯仿提取后水提剂的黄芩苷与黄芩生药比 (苷药比 )较低 (2 0 5 % ) ;而正己烷、氯仿提取液中均未测到黄芩苷。分解剂中 ,柴胡、黄芩与人参三味药组的苷药比 (5 4 5 % )略高于单味药组的苷药比 (5 38% ) ,二者均明显高于全方组苷药比 (2 36 % )。结论 :黄芩苷在各分散系中的分布随极性变化而变化 ;而小柴胡汤中柴胡、黄芩、人参三味药的配伍组合对黄芩苷似有助溶作用。
Objective:To explore the extracting methods of baicolin from ShiSaico Tong(SST) by comparing the content of baicolin in different agents.Methods:Got different extracted and devidied agents through extracting baicolin from SST with alchcol,chloroform and n-hexane,and devide SST with the prescript theory.HPLC was used to determine the content of baicolin in different agents.Results:During the extracted agents,the proportion of baicolin to drug extracted by alchcol was the highest(3.45%),the proportion of that which left after extracted with n-hexane and chloroform was the lowest (2.05%).No baicolin was determined in the extract with chloroform or n-hexane.Scutellaria decocted with bupleuri and ginsen was the best way,the proportion (5.45%) was higher than scutellaria decocted alone,significantly higher than the SST.Conclutions:Extracted with buleuri and ginsen by alchcol was the better method for the baicolin from SST.
