摘要
采用加长组合琼脂糖凝胶电泳,可以有效地分辨水稻BT型不育系(寒丰A)和保持系(寒丰B)线粒体DNA HindⅢ,BamH Ⅰ和Sal Ⅰ酶切片段的数目和分子量。寒丰不育系和保持系线粒体基因组DNA最小长度约300kb,无显著差异。用9个玉米和水稻线粒体基因探针与酶切条带杂交发现,不育系与保持系线粒体DNA的相同大小酶切片段含有相似的顺序。两种mtDNA均存在rrn18S的重复顺序。不育系与保持系线粒体DNA HindⅢ,BamH Ⅰ和Sal Ⅰ酶切产生的相同片段分别为70%,64%和71%。
The amount and molecular weight of fragments of mtDNA digested by Hind Ⅲ, BamH I, and Sal I of BT male sterile and maintainer lines can be distinguished by using agarose gel electrophoresis. Theresults showed that. 1) the minimum size was 300kb, no distinct length difference was found on mtDNAbetween MS line and maintainer; 2) Homologous sequences of mtDNA were found between MS and maintainer lines by using 9 probes of maize and rice to hybridize with digested mtDNA. The mtDNAs of MS and maintainer lines have rrn 18s repeated sequences; 3) Percentages of the homologous found in mtDNA between MS and maintainer lines digested by Hind Ⅲ, BamH I and Sal I were 70%, 64% and 71%, respectively.
