摘要
目的 观察嗅球成鞘细胞 (OECs)培养液对培养中的新生大鼠大脑皮层神经元的影响。方法 原代培养新生大鼠OECs和大脑皮层神经元 ;收集含有OECs分泌物的培养液并以不同剂量添加到正在培养的神经元中 ,观察神经元正常及损伤时的生长情况。结果 培养的OECs形态学观察及神经生长因子受体 (p75NGFR)、胶质纤维酸性蛋白 (GFAP)、胶质细胞源性营养因子 (GDNF)免疫组化阳性反应均与报道一致。OECs培养液 (0 .4~ 1.0ml)能明显促进神经元生长速度 ,并能有效地减少受损神经元的崩解。结论 OECs有促进正常神经元生长、保护损伤神经元的作用 。
Objective To investigate the effect of olfactory ensheathing cells(OECs) culture medium on culture neurons of cerebral cortex from neonatal rats.?Methods With primary culture method,OECs culture medium was gathered and added into the medium of culture cerebral cortex neurons by different doses. The growth status of neurons was observed both in normal and injury situation.?Results The morphological and immunochemical properties of OECs were consistent with literatures. Administration of OECs culture medium (0.4 1.0ml) enhanced growth of neurons and decreased degeneration of injuried neurons.?Conclusion These results suggest that OECs is capable of promoting growth of neurons and protecting neurons from death induced by injury,these effect may owe to the bioactive substance secreted by OECs.
