汉防己甲素对成纤维细胞游离Ca^(2+)浓度影响的机制探讨 被引量:4

摘要 目的 探讨汉防己甲素 (Tet)对成纤维细胞内游离Ca2 + 浓度影响的机制。方法 体外培养人胚肺成纤维细胞 (HLF) ,应用Fura_2 /AM荧光法 ,观察Tet对HLF游离Ca2 + 浓度的影响 ;同时应用比色法 ,测定细胞培养液一氧化氮 (NO)水平 ;研究Tet对HLF游离Ca2 + 浓度的影响与NO间的关系。结果 与正常对照组比较 ,Tet能明显降低HLF游离Ca2 + 浓度 ,同时明显升高细胞培养液NO水平。结论 Tet对成纤维细胞的钙拮抗作用与其增加NO合成。 Objective To study the mechanism of Tetrandrine (Tet) on the effect of the free calcium co ncentration inside of fibroblast, and the relation between the role of Tet on the free calcium co ncentration inside of fibroblast. Methods Culture human embroyo lung fibroblast(HLF) in vitro.The methods of Fura_2/AM fluorescence and colorimetry were used to observe the role of Tet on the free c alcium concentration inside of fibroblast and NO level of cell culture fluid r espectively. ResultsCompared with the normal control group,Tet could si gnific intly decrease the free calcium concentration inside of fibroblast and increase NO level of cell culture fluid. Conclusion The effect of Tet on the free calcium concentration inside of fibroblasts is associated with NO synthesis and release.
