冷冻对视网膜色素上皮细胞分泌肝细胞生长因子的影响

摘要 本研究观察冷冻处理后的人视网膜色素上皮细胞(human retinal pigment epitheliumcells,hRPE)在体外培养和体内玻璃体环境中分泌肝细胞生长因子(hepatocyte growth factor,HGF)水平的变化,调查变化后的玻璃体对正常RPE细胞的促增生作用。体外培养的RPE细胞在-80℃下进行冷冻,冷冻时间分为0s、15s和60s,随后继续体外培养和注入正常兔眼玻璃体,冷冻后的第3d和6d收集细胞培养液和玻璃体样本,ELISA法测定hHGF含量;进一步,用MTT法测定正常RPE细胞加入玻璃体样本后的生长状态。结果显示冷冻刺激RPE细胞hHGF的分泌增多,并可促进RPE细胞增生。 To investigate hepatocyte growth factor (HGF) concentration in frozen human retinal pigment epithelium cells (hRPE) both in vitro and in vivo. To evaluate the effect of HGF induced hRPE proliferation. Cultured hRPE cells (3rd-5th passages) were frozen at - 80℃ for 15 seconds and 60 seconds. The following substances were tested: (1) The frozen cells were incubated for further. (2) Intravitreous injection of frozen cells in Rabbits. The cell supernate and vitreous samples were collected at 3 days and 6 days. The concentration of HGF was monitored by ELISA assay. (3) Some vitreous samples were added into non-freezing hRPE cells culture media and the cell proliferation was examined by MTT assay. We found that freezing can increase expression of HGF in hRPE cells. The vitreous samples (with frozen hRPE cells) can induce the hRPE cell proliferation.
