摘要
目的 :研究初发急性白血病 (AL)患者骨髓、外周血单个核细胞端粒酶活性及端粒酶逆转录酶基因(hTERT)的表达 ,探讨其临床意义。方法 :采用端粒酶PCR ELISA法检测 2 4例初治AL患者的端粒酶活性 ;采用逆转录 聚合酶链反应 (RT PCR)方法检测hTERT基因mRNA的表达水平。结果 :在 2 4例初发AL患者骨髓、外周血样本中 ,18例 (75 .0 % )表达端粒酶活性 ,其吸光度 (A )均值分别为 0 .5 38± 0 .0 6 2和 0 .4 6 3± 0 .0 5 4 ,而 12例正常人外周血中只有 1例表达端粒酶活性 A 均值为 0 .16± 0 .0 12 ,前二者分别与后者比较差异有极显著性意义 (P <0 .0 1)。在 2 4例AL患者中 ,19例 (79.2 % )表达hTERT基因mRNA。 2 4例AL骨髓样本中 ,17例既有端粒酶活性表达 ,又有hTERT基因mRNA表达 ,端粒酶活性的表达和hTERT基因mRNA的表达具有明显的一致性。结论 :初发AL患者骨髓、外周血细胞端粒酶活性及hTERT基因mRNA表达明显高于正常 ;AL患者细胞端粒酶活化与其hTERT表达密切相关。
Objective:To investigate telomerase activity and human telomerase reverse transcriptase (hTERT) expression in acute leukemia and their clinical signifcance.Method:Telomerase activity was examined by polymerase chain reaction enzyme-linked immunoassay (PCR-ELISA). Expression of hTERT was detected by reverse transcription-polymerase chain reaction (RT-PCR) in 24 cases of acute leukemia and in 12 cases of normal human peripheral blood.Result:In the bone marrow and peripheral blood of 24 acute leukemia, telomerase activity was detected in 75.0% for both in which the absorbance (A) were 0.538± 0.062, 0.463± 0.054,respectively. Whereas in 12 normal peripheral blood, telomerase activity was 0.16± 0.012, only a positive rate of 8.3%.Telomerase activity in the bone marrow and peripheral blood of acute leukemia were significantly higher than that in normal control(P< 0.05). RT-PCR analysis revealed that hTERT mRNA was expressed in 79.2%( 19/24) acute leukemia, but in only 1 of 12 normal peripheral blood. In 24 acute leukemia, 17 cases were all telomerase-positive and expressed hTERT mRNA. The expression of hTERT mRNA is correlated with telomerase activity(P< 0.01).Conclusion:Both telomerase and hTERT mRNA were markedly higher in acute leukemia than those in normal control. It is further proved that hTERT gene expression was strongly associated with telomerase activity in acute leukemia.
