摘要
设计针对端粒酶的HDV核酶,观察其对端粒酶活性抑制。设计和合成了针对端粒酶组分的HDV核酶的序列,构建了该核酶的体外转录和真核表达质粒,检测了HDV核酶对端粒酶组分的体外切割效力和对端粒酶活性的抑制作用。核酶对底物的最大剪切效率为70.4%。导入细胞后,使端粒酶活性下降90%。HDV核酶(g.RZ57)对端粒酶活性有抑制作用,可望成为有效的肿瘤治疗抑制剂。
To design HDV ribozyme for human telomerase RNA(hTR) component, and observe its inhibition for telomerase activity. Pseudoknotted g.RZ57 was designed and synthesized. In vivo transcription plasmid and eukaryotic expression plasmid of ribozyme were constructed. Then we determine the cleavage activity of ribozyme and detect telomerase activity continuously. HDV ribozyme showed a specific cleavage activity against the telomerase RNA. The telomerase activity dropped to 10% of that before. HDV ribozyme(g.RZ57) can be potentially used as inhibitor for cancer therapy.
