摘要
目的 :探讨伊贝沙坦 (Irb)对血管平滑肌细胞 (VSMC)分泌细胞因子的影响。方法 :兔 VSMC分 6组处理 ,即对照组 ,低密度脂蛋白组 ,氧化低密度脂蛋白 (ox L DL)组 ,Irb不同浓度组 ,Irb不同时间组及 Irb+ox L DL 组。以酶联免疫法检测培养上清液中细胞因子 TNFα,IL- 6 ,IL- 8水平。结果 :ox L DL(5 0 m g/L)对 TNFα,IL- 6 ,IL- 8分泌水平于刺激后 8h增加至 6 .4~ 8.5倍 ;Irb呈浓度 (0 .0 1~ 10 0 μmol/L)、时间 (1~ 2 4 h)依赖地抑制基础及 ox L DL 刺激的细胞因子分泌 (与对照组比较均 P<0 .0 5 )。结论 :Irb可能通过抑制基础及 ox L DL
AIM:To investigate the effects of irbesartan(Irb) on cytokins secretion by vascular smooth muscle cells(VSMC). METHODS:Rabbit aortic VSMCs were cultured in 6 groups: control,the low density lipoprotein,the oxidized low density lipoprotein(oxLDL),the Irb+oxLDL and the Irb of different concentrations and time groups. The level of tumor necrosis factor alpha(TNFα),interleukin-6(IL-6) and interleukin-8(IL-8) were evaluated by ELISA method.RESULTS:oxLDL increased the secretion of TNFα,IL-6 and IL-8 by 6.4~8.5-fold in 8 h. Irb decreased the cytockins secretion in a concentration(0.01~100 μmol/L)-and time(1~24 h)-dependent manner,and inhibited the above effects of oxLDL. CONCLUSION:Irb could regulate the biological effects of VSMC by inhibiting base and oxLDL-stimulated cytokins secretion.
