摘要
为了解不同种植期棚室土壤放线菌种群变化趋势,采用高通量测序技术分析土壤放线菌群落的变化,探讨作物种植期对土壤放线菌群落结构产生的影响。土壤理化分析结果表明定植期、坐果期及盛果期土壤的可溶性盐含量显著高于拉秧期,p H则显著低于拉秧期。拉秧期可培养放线菌数显著高于其他三期。高通量测序结果显示4样本共测47000条有效序列,分属于133个OTU,其中定植期78个,坐果期61个,盛果期79个,拉秧期70个。多样性指数分析、Bata多样性分析及热图分析均显示拉秧期土壤放线菌群落与其他三组有所不同。群落组成分析显示4个种植期土壤样本中链霉菌占约30%以上,分支杆菌和棒状杆菌分别占比超20%,是土壤放线菌的绝对优势菌群。无枝酸菌属(Amycolatopsis)、迪茨氏菌属(Dietzia)、气微菌属(Aeromicrobium)、节细菌属(Arthrobacter)比例发生显著变化可能与种植作业过程中化肥、农药等农业操作有关。
In order to know the differenceofsoil actinomycetes communities during growth stages in greenhouse,high-throughput sequencing technology wasintroduced.Physical and chemical analyses showed that the content of soil soluble salt was significantly higher at planting,bearingfruit and productive stages than at uproot stage,but pH value was lower significantly.Culturable actinomycetequantity was significantly higher at uproot stage than atthe other three stages.The 47000 effective sequences acquiredwithhigh throughput sequencing belonged to 133 OTU,including 78,61,79 and 70 OTU,respectively,at planting,bear fruit productive and uproot stages.Diversity index,bata analysis and heatmap analysis showed that soil actinomycete community was different betweenthe uproot stage and the other three stages.The community composition analysis showed that more than 30%were Streptomyces,more than 20%were Mycobacterium and Corynebacterium in all soil sample,which were dominant groupsof soil actinomycetes.The proportion of arthrobacter,Amycolatopsis,Dietzia,and Aeromicrobium wassignificantly changed at uproot stage,which may be correlated with fertilizers andpesticides during cultivation period.
