摘要
应用聚合酶链反应 (PL-PCR)技术和噬菌体裂解试验同时对 80株鼠疫菌 ,1株假结核株、 1株大肠杆菌埃希氏菌进行检测并作比较。结果显示经噬菌体裂解试验 82株菌均在 18~ 2 0 h显示出噬菌斑 ,而其中 2株聚合酶链反应为阴性 ,另 80株全部为鼠疫菌特异性扩增带 ,比较清晰、典型、稳定 。
Amplification of Y. pestis DNAs has been proposed as a detection method. A PCR assay was compared with the test of Bacteriophages Lysis by detection of eighty strains of Y. pestis, one straint. Y. pseudotuberculosis and one strain of Escherichia coli. The results show that all strains of Y. pestis appeared the phage plagues in 18 to 20 h. Y. pseudotuberculosis and E. coli did as well. DNA from eighty strains of Y. pestis were amplified and producted a PCR product which was the same as the predicted size. No amplifications were detected in Y. pseudotuberculosis and E coli. All results suggest that the PCR assay is a specific? sensitive and rapid method for detection of Y. pestis, providing a useful tool for epidemiological and surveillance studies of plague.\;
