摘要
目的 通过比较艾灸不同腧穴对痴呆(Alzheimer disease,AD)大鼠血清中瘦素(leptin),脂肪组织中瘦素、瘦素受体(leptin receptor)水平的表达,探讨艾灸神阙、足三里穴对AD大鼠脂质代谢的影响。方法 在55只大鼠中先随机抽取10只为空白对照组,剩余45只在造模成功后筛选出40只随机分为模型对照组、神阙组、足三里组、神阙-足三里组,每组10只。大鼠腹腔注射D-半乳糖、氢溴酸东莨菪碱制造痴呆模型,各治疗组艾炷灸相应腧穴,每穴3壮。用酶联免疫吸附法检测大鼠血清中瘦素的含量,用荧光定量RT-PCR法检测大鼠脂肪组织中瘦素mRNA的表达,用蛋白质免疫印迹法(Western blot)检测大鼠脂肪组织中瘦素受体蛋白的表达。结果 与模型组比较,各艾灸组大鼠血清中瘦素含量明显降低(P<0.01),瘦素受体含量明显增多(P<0.01),瘦素基因含量明显增多(P<0.05),各治疗组之间比较差异不明显(P>0.05)。结论 艾灸神阙、足三里穴均可提高血清中瘦素、脂肪组织中瘦素及瘦素受体水平。临床治疗AD可参照本实验研究结果选穴。
Objective Giving moxibustion to different experimental acupoints and made comparison to leptin,leptin in adipose tissue and leptin receptor in rats serum expression to discuss the metabolism effect by moxibustion Shen Que(RN 8)point and Zu San Li(ST 36)point.Methods Random drawing 10 rats as a normal group from 55 rats,then after successfully established experiment,40 rats models were selected and divided as model group,Shen Que(RN 8)point group,Zu San Li(ST 36)point group and Shen Que-Zu San Li(RN 8-ST 36)group with 10 rats in each group.Rats were intraperitoneally infused with D-Gal,scopolamine hydrobromide to made the dementia model,and gave moxibustion treatment to three treatment group,each acupoint 3 cones,the leptin content were analyzed by ELISA method,using fluorescence quantitative RT-PCR to detect the genes expression of rats adipose tissue and detect the leptin receptor protein with Western Blot.Results The leptin concentration was decreased clearly(P<0.01)in every model groups and the content of leptin gene(P<0.05)and leptin receptor(P<0.01)was increased significantly.The curative effect of every treatment group were the same.Conclusion It could improve the leptin in serum,leptin in adipose tissue and leptin receptor level by giving moxibustion to Shen Que(RN 8)point,Zu San Li(ST 36)point and the clinical treatment of AD can refer to the results of this research.
