补肾抗癌方对TAM诱导的小鼠Lewis肺癌中VEGF表达影响 被引量:4

摘要 目的探讨补肾抗癌方对不同活化表型的肿瘤相关巨噬细胞(TAM)诱导的肺腺癌小鼠移植瘤模型中血管内皮生长因子(VEGF)表达的影响。方法构建不同活化表型TAM的肺腺癌小鼠移植瘤模型,将50只C57BL/6J小鼠随机分成5组:空白组;TAM模型对照组;TAM中药组;活化TAM模型对照组;活化TAM中药组。于接种后给予相应药物干预,第21天处死各组小鼠,剥离皮下肿瘤,流式细胞仪检测TAM的数量;采用免疫组织化学法检测小鼠皮下移植瘤组织中VEGF表达。结果在建模第21天,各中药组小鼠移植瘤体积明显小于各模型对照组,TAM模型对照组小鼠移植瘤体积明显小于活化TAM模型对照组,各中药组小鼠移植瘤组织中CD206表达水平显著低于各模型对照组,各中药组小鼠移植瘤组织中VEGF表达水平低于各模型对照组。结论补肾抗癌方可能通过抑制TAM和VEGF表达从而达到抗肿瘤的效果。 Objective To study the tonifying kidney and anticancer prescription on the vascular endothelial growth factor( VEGF) expression in different activation phenotype of tumor-associated macrophages( TAM) induced lung adenocarcinoma xenograft mouse model. Methods Different TAM activation phenotype in mice lung adenocarcinoma xenograft model were constructed,and 50 C57 BL /6J mice randomly divided into 5 groups: control group,TAM model control group,3. TAM TCM group,activated TAM model control group,and activated TAM TCM group; appropriate drug intervention was applied after inoculation,all mice were executed on day 21,subcutaneous cancer was peeled,the number of TAM was calculated by flow cytometry; and immunohistochemistry staining method was used to detect the expression of VEGF subcutaneously transplanted tumor tissue in mice. Results On day 21 after modeling,volume of the TCM mouse xenograft was significantly smaller than that of the control group,volume of TAM mouse xenograft model control group was significantly smaller than the activation TAM model control group,CD206 cells ratio of mice transplanted tumor tissue in the TCM group was significantly lower than the proportion of each model control group,the expression of CD206 in tumor tissues of TCM group was lower than the models in other groups. Conclusion Tonifying kidney and anticancer prescription may inhibit the expression of TAM and VEGF to achieve antitumor effect.
