摘要
目的:研究Foxp3基因在CD4+、CD25+调节性T细胞(Treg细胞)中的表达水平及其对Treg细胞的调节作用。方法:应用流式细胞术(FACS)测定实验组和对照组患者术前外周血中T淋巴细胞及Treg细胞含量的改变;采用RT-PCR技术检测FoxP3基因在胃癌患者外周血Treg细胞中的表达与对照组的差异。结果:胃癌患者外周血中T淋巴细胞(CD3+T)及抑制杀伤性T细胞(CD8+T)总数,与健康对照组相比无明显统计学差异(P>0.05);Treg细胞含量显著高于健康对照组(P<0.01);FoxP3基因在胃癌患者外周血Treg细胞中呈现高表达,明显高于健康对照组(P<0.01)。结论:FoxP3基因可能与Treg细胞发挥抑制胃癌病人产生的抗肿瘤自身免疫功能作用有着密切的关系。
Objective:To investigate expression of FoxP3 gene in Treg cells and the possible function in Treg cells in regulating anti-cancer response in the gastric carcinoma patients.Methods:By using flow cytometry, the levels of CD3+T cells, CD8+T cells, and Treg cells in peripheral blood of preoperative patients with gastric cancer were detected respectively, and the results were analyzed statistically in comparison with that of the healthy volunteers;RT-PCR techniques were used to detect the expression of FoxP3 gene in these Treg cells, and the results were analyzed statistically in comparison with that of the healthy volunteers.Results:The percentages of total T lymphocytes (CD3+T) and inhibiting killer T cells (CD8+T) in peripheral blood of the preoperative patients with gastric cancer were found to be no different statistically in comparison with that of healthy volunteers(P>0.05); the percentage of Treg in total CD4+T cells in patients with gastric cancer increased significantly compared with that in volunteers (P<0.01); The results of the FoxP3 gene expression on Treg cells in the patients with gastric cancer were significantly higher than that in healthy volunteers (P<0.01).Conclusion:Increased FoxP3 gene expression of Treg cells indicates that FoxP3 genes may play a pivotal role in regulating the function of Treg cells in self-tumor immunity of the patients with gastric cancer.
