分装容器对培养基热穿透、灭菌效果及灵敏度的影响

摘要 目的分析不同分装容器内培养基的热穿透、灭菌效果、灵敏度差异以及三者之间的相互关系。方法将硫乙醇酸盐流体培养基、胰酪大豆胨液体培养基分装于中试管和血浆瓶,用TMI温度探头比较两种容器的热穿透差异,用灭菌指示剂比较不同容器中培养基灭菌效果,灵敏度试验比较不同容器中培养基灵敏度。结果血浆瓶热穿透力<中试管热穿透力;两种容器内培养基灭菌效果和灵敏度结果均无差异。结论 116℃蒸汽灭菌30 min不会破坏硫乙醇酸盐流体培养基、胰酪大豆胨液体培养基的有效营养成分,且能达到灭菌目的;两种培养基的热穿透性因分装容器种类而异,但灭菌效果、灵敏度与分装容器无直接关系。 Objective To investigate the difference and relationship of heat penetration,sterilization effect and sensitivity of culture medium in different containers.Methods Fluid thioglycolate medium and tryptic soy broth medium were infused into tubes and sealed bottles,of which the heat penetrations were compared by TMI temperature recorder,while the sterilization effects by sterilization indicator,and the sensitivities by sensitivity test.Results The heat penetration ability of sealed bottles was lower than that of tubes.However,no differences were observed in the sterilization effect and sensitivities of media in the two containers.Conclusion Sterilization at 116℃for 30 min could not destroy the nutrients of fluid thioglycolate medium and tryptic soy broth medium.The heat penetrations of two media in different containers were also different,while the sterilization effects and sensities showed no significant difference.
