摘要
在单因素试验基础上进行部分因子试验和中心点组合试验,优化蚕蛹蛋白的酶解条件.结果表明,最佳酶解条件为:温度45℃,加酶量2 000U/g,酶解时间4h,蛋白/水1∶9.19,pH值2.71;通过3次试验验证,蚕蛹蛋白平均水解度为57.08%,与预测最大值的误差为0.51%.在最优的酶解条件下能准确地获得理想的水解度.
In order to optimize the enzymatic hydrolysis condition of acid protease of silkworm pupae protein,based on the single-factor experiment a quadratic-multinomial-mathematical model is established by Response Surface Methodology(RSM).By the center point design experiment,the optimized hydrolysis conditions are determined to be:temperature of 45℃,E=2 000U/g,hydrolysis time of 5hours,protein/water=1∶9.19,pH=2.71.From the three experiments, the average degree of hydrolysis of silkworm pupae protein is 57.08%,the experiment error is 0.51%,and the DH is ideal by the optimized design.
