摘要
建立了超高效合相色谱(Ultra Performance Convergence Chromatography, UPC2)法同时测定奶粉中β-胡萝卜素和叶黄素的方法。超高效合相色谱(UPC2)技术集合超临界流体色谱(Supercritical Fluid Chromatography, SFC)和超高效液相色谱(Ultra Performance Liquid Chromatography, UPLCTM)的技术优点,流动相CO2为主体,乙醇/乙腈(1︰1)为助溶剂。选用ACQUITY UPC2 BEH 2-EP色谱柱,流速为0.8 mL/min,检测波长为438 nm,上述2种类胡萝卜素在0.025 6~2.636 8μg/m L范围内有良好的线性关系,相关系数R2均大于0.998,在0.8~1.8 mg/kg范围内平均回收率在89.8%~98.7%之间,每个加标水平的6次重复试验RSD<7.8%。试验表明该方法简便、快速、准确,可同时测定乳品中的叶黄素和β-胡萝卜素含量。
A new method was developed for the determination of lutein andβ-carotene in milk products by Ultra Performance Convergence Chromatography(UPC2).The UPC2 was mixture with Supercritical Fluid Chromatography and Ultra Performance Liquid Chromatography,mobile phase CO2 as the main body,ethanol/acetonitrile(1︰1)as the cosolvent,methanol at a flow rate of 0.8 mL/min.Using the ACQUITY UPC2 BEH 2-EP column,the UV detector was set at a wavelength of 438 nm.The linearities for two carotenoids were good at concentration ranges of 0.025 6-2.636 8μg/mL,and the correlation coefficients were greater than 0.998.The average spiked recoveries for three addition levels were from 89.8%to 98.7%.The relative standard deviation of each spiked test was less than 7.8%.The method could simultaneously determine lutein andβ-carotene levels in powdered milk with simpleness,speediness and accuracy.
