摘要
目的评定高效液相色谱法测定肌酸粉中一水肌酸的不确定度。方法肌酸粉中一水肌酸经过纯化水超声提取、色谱柱分离、流动相等度洗脱,使用紫外检测器波长为220 nm检测,外标法定量测定肌酸粉中一水肌酸的含量。根据JJF1059.1-2012《测量不确定度评定与表示》,考察称量、标准溶液配制、样品和仪器重复测量等主要因素引入的不确定度,幵对不确定度各分量迚行计算和合成。结果当肌酸粉中一水肌酸含量为85.9%时,在95%的置信区间下,其扩展不确定度为0.276%(k=2)。评定结果表明,实验过程中的不确定度,主要来源于仪器测量的重复性和样品处理过程的引入。结论此不确定度评定结论适用于高效液相色谱法测定肌酸粉中一水肌酸含量的不确定度分析,对检测结果的准确度提高具有一定的指导意义。
Objective To evaluate the uncertainty of determination of creatine monohydrate in creatine powder by high performance liquid chromatography.Methods The creatine monohydrate in the creatine powder was ultrasonically extracted by purified water,separated by column chromatography,and e luted with equal flow.The ultraviolet detector was used to detect the samples with the wavelength of 220 nm.The content of creatine monohydrate in creatine powder was determined by external standard method.According to JJF 1059.1-2012 Measurement Uncertainty Evaluation and Representation,the uncertainty introduced by the main factors such as weighing,standard solution preparation,sample and instrument repeated measurement were investigated,and the components of uncertainty were calculated and synthesized.Results When the creatine content of creatine powder was 85.9%,the extended uncertainty was 0.276%(k=2)under the 95%confidence interval.The evaluation results showed that the uncertainty during the experiment mainly came from the repeatability of instrument measurement and the introduction of sample processing.Conclusion The uncertainty evaluation conclusion is applicable to the uncertainty analysis of the determination of creatine content in creatine powder by high performance liquid chromatography,which has a guiding significance for improving the accuracy of the test results.
