摘要 目的 :观察脐血清能否替代正常人AB血清用于人PBMC体外诱导CIK的细胞培养过程。方法 :①按照本室的方法体外分别用含正常人AB血清或脐血清的培养基诱导正常人 (脐血组和正常组各 10例 )CIK ,比较不同血清诱导CIK的扩增差异 ;②流式细胞仪检测CIK细胞表面标志CD3、CD56;③ 3H -TdR释放法检测CIK对肝癌细胞系SMMC -772 1、Bel-740 2、Hep -3b ,肠癌细胞系HIC -2 51,正常胎肝细胞系L -0 2 ,红白血病细胞系K562及经阿霉素诱导的耐药K562细胞毒作用。结果 :①不同血清诱导的CIK在细胞扩增曲线及细胞表面标志上无差异 ,细胞扩增倍数达 64.3倍和 67.4倍 (t =1.0 0 3 ,P >0 .0 5) ,CD3 + CD56+ 细胞扩增倍数达 80 0倍以上 ;②脐血组CIK对肝癌细胞SMMC -772 1、Bel-740 2、Hep -3b、HIC -2 51杀伤能力达 70 %~ 80 % ,与正常组相同 ,杀伤能力无差异 ;③对正常胎肝细胞系L -0 2的细胞毒作用 <5% ;④二组CIK对已诱导耐药的K 562细胞和未诱导耐药的K562细胞细胞毒作用均达到 70 %左右。结论 :脐血清组和正常人组CIK一样对肝癌细胞有很强的细胞毒作用 ,对正常肝细胞无损伤 ,并且对耐药肿瘤同样有效 。 Objective To observe if the umbilic al sera could replace normal AB sera in the processes that the CIK be induced from PBMC. Methods ①Inducing CIK respectively in vitro from P BMC of normal people with cultures containing normal AB sera or umbilical sera, to find out whether there is any difference between two groups of CIK while the cells increasing; ②detecting the cell surface markers CD3 and CD56 of two group s; ③detecting cytotoxicity of two CIK cells by 3 H -TdR release method to different target cells (SMMC-7721, Bel-7402, Hep-3b, all from human liver cancer , HIC-251, from intestine cancer, K562, leukaemia, and L-02, from fetal liver bu t not cancer). Results the number of cells of PBMC increas ed both in normal sera culture and in umbilical sera culture as vigorously as mo re than 60 times(t=1.003,P>0.05), and the CD3 +CD56 + cells, above 800 times; The cytotoxicy both groups cells to the cell lines of hepatocellular car cinoma were as high as 65%~81%; two CIK could also give a high cytotoxicity abo u t 72% to the K562 cell line which expressed multi-drug resistance. But it had no effect on L-02, the fetal liver cell line. Conclusion The CIK induced in the culture containing umbilical sera possess all capability as normal group such as vigorous increase in number and strong cytotoxicity to t umor cell in vitro and indeed harmless to normal cell. It should be useful in cl inical apply and deserves further studies.
