实验性肝纤维化过程中基质分解素-1酶原的表达变化

摘要 目的 研究实验性肝纤维化过程中基质分解素 -1(MMP -3 )酶原蛋白表达的动态变化。方法 建立四氯化碳中毒性大鼠肝纤维化模型 ,采用半定量Western免疫印迹方法 ,对肝纤维化不同阶段肝内MMP -3的酶原蛋白的表达水平进行了检测。结果 在正常大鼠肝组织中有MMP -3的酶原表达。在肝纤维化发生发展过程中MMP -3酶原蛋白表达水平逐渐增强 ,至肝纤维化中期到达最高峰 ,到肝纤维化晚期及肝硬化阶段其表达较中期下降 ,但仍高于正常对照水平。结论 MMP -3在肝纤维化的发生。 Objective To investigate the dynamic changes of stromelysin-1(metalloproteinase-3, MMP-3)proenzyme protein expression in the process of experimental liver fibrosis.Methods The carbon tetrachloride-induced hepatic fibrosis rat model was used for the study. MMP-3 proenzyme protein expression in different stages of liver fibrosis was measured by quantitative analysis of Western blot.Results MMP-3 proenzyme protein could be detected in normal rat liver. During the development of liver fibrosis, MMP-3 proenzyme protein level increased gradually,with the expression being maximum at the intermediate stage of fibrosis. Its expression decreased gradually during the late liver fibrosis and at the stage of liver cirrhosis, but remained relatively high above normal. Conclusion MMP-3 may play an important role in the pathogenesis of liver fibrosis and cirrhosis.
