目的 探讨银染法在三链形成的电泳迁移率改变分析 (EMSA)中的可行性 ,从而为三链核酸研究提供有效的非同位素检测方法。 方法 三链形成反应后进行 EMSA分析 ,然后用银染显示电泳形成的 DNA区带。 结果 银染法显示出三链的 EMSA滞后带 ,并揭示三链形成的平台效应。 结论 本文首次表明银染法可用于三链形成的 EMSA分析快速检测 ,且 SOX9基因内含子
Objective To apply silver staining in detection of altered EMSA bands of stable triple-helix DNA,thus to introduce a non-isotopic method for rapid and simple detection of triplex formation. Methods Show the separated DNA bands created by their differential electrophoresis rates in EMSA by silver staining. Result Silver staining reveal the lower electrophoresis rates of triplex as well as the plateau phase of triple-helix DNA formation. Conclusion This is the first presentation of non-isotopic detection of triplex by silver staining, and also triplex formation in the first intron of human SOX9 gene.
