摘要
目的 :观察兔心肌缺血再灌注 (I/R)损伤中内皮功能改变 ,参麦注射液 (SMI)对其影响及作用机制。方法 :检测假手术对照组、心肌I/R模型组及心肌I/R加SMI治疗组 ,不同时期血中一氧化氮代谢产物 (NOP)和内皮素 (ET)含量及心肌组织NOP、ET、总超氧化物歧化酶 (T SOD)和丙二醛 (MDA)含量 ,并电镜观察心肌超微结构。结果 :I/R模型组与假手术对照组比较缺血 4 0min、再灌注 4 0min血清NOP明显降低 ,血浆ET显著增高 ;再灌注 4 0min后心肌组织NOP、T SOD明显降低 ,ET、MDA明显增高 ,心肌超微结构发生异常改变。而I/R加SMI治疗组与I/R模型组比较上述改变减轻。结论 :心肌I/R导致血管内皮功能紊乱 ;SMI能够改善I/R时的内皮功能 。
Objective:To study the change of endothelial function during myocardial ischemia and reperfusion (I/R) injury of rabbits, the effect of Shenmai Injection (SMI). Methods:Twenty one Japanese white rabbits were randomly divided into 3 groups (n=7 each): sham operated group (A), I/R group(B), and SMI treated I/R group (C). The concentration of serumal nitric oxide production (NOP) and plasma endothelin (ET) before ligation of the coronary artery, 40 min after ischemia and 40 min after reperfusion were measured. The concentration of NOP, ET, total superoxide dismutase (T SOD)and malonyl dialdehyde (MDA) of myocardial tissue were also measured. The ultrastruction structure of myocardial cell was observed by electro microscope. Results:① Compared with group A, serumal NOP in group B decreased and plasma ET increase at 40 min after ischemia (P< 0.01 ), 40 min after reperfusion. 40 min after reperfusion, NOP and T SOD in group B were lower significantly than group A, ET and MDA were higher significantly (P< 0.01 ). Ultrastructure of myocardial cell was abnormal. ②SMI can increase serumal NOP in 40min after ischemia and 40 min after reperfusion and lower plasma ET in 40min after reperfusion significantly (P< 0.01 ). SMI can weaken the abnormality of group B. Conclusion:Myocardial I/R leads to abnormality of vascular endothelium function, SMI can improve endotheliual function and reduce myocardial I/R injury.
