摘要
运用Bal 31末端缺失技术将人IL-2Rα cDNA进行3’—5’的单向缺失,获得了IL-2RαcDNA3’端的系列缺失子。经DNA顺序分析表明:这些缺失克隆分别含有不同位置上的Poly(A)信号序列,将它们克隆到穿梭质粒P^(91023)上,得到了系列缺失子的表达性质粒P^(91-IL2R),为进一步分析Poly(A)及3’非转译区(3’UTR)对IL-2Rα基因转译调控的影响创造了必要条件。
Series mutants of human IL-2Rα cDNA in 3' end were produced with exonuclease-Bal 31. Sequencing analysis demonstrated that the mutants contain poly (A) signal sequence at different region,respectively. They were subcloned into a shuttle plasmid and several expressive plasmids-P^(91-IL2R) were obtaind . It is very essential for the further study of Poly (A) and 3'-untranslational region(3'UTR) control the translation of IL-2Rα gene.
