摘要
本文报导了应用HIV-1病毒裂介物免疫Balb/c小鼠,采用杂交瘤技术制备了大量单抗,继用FACS、免疫印染法(Western Blot;WB)、组织切片免疫酶染色法及不同HIV-1 ELISA试剂盒进行单抗特异性鉴定。确定了48个特异性抗HIV-1结构基因表达的蛋白成分的单抗;同时也发现了由于免疫抗原不纯,一般市售HIV-1抗体筛选用ELISA试剂盒及WB试剂盒存在着污染的人细胞成份,故使制备的单抗中很大一部分为抗污染的人细胞蛋白成分的非特异性抗体。
Forty-eight monoclonal antibodies (McAbs) were raised against human immunodeficiency virus (HIV-1) lysate and characterized by ELISA, western blot immunohistological stainning .They recognized different HIV-1 structural proteins p55 +
p24p,55+p18, p55 + p24 + 130......, p24, p18, p18 + p27, gpl60+gp120 and gpl60+gpl20+gp41.
It has been demonstrated that the HIV-1 ysate, commerial HIV-1 ELISA kit westrn blot kit was contaminated by the proteins or human T cell line. The experience showed that immunohistological or FACS examination of hybridomas supernatant was necessary to eliminate nonspecific antibodies before the were cloned.
