脑梗死后细胞骨架MAP-2、NF200与Bcl-2的相关性研究 被引量:1

摘要 目的 探讨人脑梗死后神经元和轴突的细胞骨架与细胞凋亡的相关性。方法 应用 10例脑梗死后的尸检标本 ,常规HE染色和免疫组织化学染色 (SP)检测微管相关蛋白 (MAP 2 )、分子量为 2 0万的神经纤维中间丝(NF2 0 0 )、凋亡相关基因 (Bcl 2 ) ,进行定性和定量分析。结果 脑梗死后以梗死灶为中心 ,按照组织损伤程度由内到外分 4个区 (0~ 3区 ) ;MAP 2在 0区表达消失 ,在 1区和 2区表达增强并形态异常 ,在 3区未见明显改变 ;NF2 0 0在 0区表达消失 ,在 1区可见表达 ,在 2区表达明显并变形 ,在 3区无变化 ;Bcl 2在 0区和 1区表达很少 ,在 2区和3区表达最显著。结论 人脑梗死后涉及细胞骨架改变和细胞凋亡的发生 ,两者呈正相关。对此过程中涉及到的因素进行干预 ,可能改变卒中的进程和预后。 Objective To investigate the relationship between cytoskeleton and apoptosis in ischemic zone of human infarcted brains.Methods We used HE and immunohistochemical methods to define changes of cells in post mortem specimens from ten patients.Microtubule associated protein 2 (MAP 2),neurointermediate filament 200 (NF 200) and anti apoptotic protein Bcl 2 were stained as markers.Results Each section of the infarcted brain was divided into four areas (the area 0~3).Immunoreactivities for MAP 2 and NF 200 were disappeared in the area 0,and preserved in the area 3.MAP 2 was increased in the area 1 and 2 markedly; however,NF 200 was increased in the area 1 distinctively and enhanced in the area 2 mildly.Bcl 2 was lost in the area 0 and 1,but increased in the area 2. Conclusion Changes of cytoskeleton and apoptosis that occur after ischemia may have a positive relationship with each other.
