摘要 目的 :观察机械牵张作用对牙周膜成纤维细胞 (PDLFs)增殖能力的影响。方法 :用自行研制的细胞加载系统对PDLFs施以频率为 6次 /min(每次为 5s牵拉、5s松弛 ) ,幅度 12 %的牵张力 ,于加载 2 4、48、96h后 ,通过细胞计数、流式细胞仪检测观察PDLFs增殖能力的改变情况。结果 :在不同的时间点 ,加力组及对照组的细胞数均不断增加 ,在 48h及 96h时加力组结果明显高于对照组。流式细胞仪结果显示在不同时间点加力组处于DNA合成期的细胞数明显高于对照组。结论 :在对培养的人PDLFs施加频率为 6次 /min、幅度 12 %的牵张力时 。 Objective: To observe the proliferation of periodontal ligament fibroblasts(PDLFs) under mechanical stretching. Methods: PDLFs were stretched by 6 cycles/min(5 seconds stretching and 5 seconds relaxing) with 12% stretching force through self-produced cultured cell loading system. At experimental time point 24,48 and 96 hours, the cells were counted and flow cytometry was employed to observe the proliferation of PDLFs. Results: 48 h and 96 h after stretch treatment the cells in experimental groups were significantly more than those in control groups respectively (P<0.05). The result of flow cytometry showed that S phase cells in experimental groups were much more than those in control groups. Conclusion: PDLFs proliferation can be promoted by proper stretch treatment.
