摘要
基于网络结构分析法研究疾病敏感基因方法是具有重要理论与实际意义的课题。本文分别对有无肾透明细胞癌的肾组织的基因表达谱做秩和检验,筛出表达差异较大的基因作为候选基因。而后对筛选得到的基因构建互信息网,基于网络结构的7个统计量,提出了"面差"的新概念,进而提出一种新的筛选可能对疾病发展过程产生重大影响的结构性关键基因的方法,并且由此得到了8个基因作为肾透明细胞癌的潜在性致病关键基因。癌症的实验研究表明得到的8个基因中有6个是与肾细胞透明癌的形成与发展有关,并且预测剩余的2个基因可能对肾透明细胞癌的产生和发展有影响。
Genes achieve their function through complex interactions in the gene network.Because the network structure determines the network function,the establishment of the genetic network of the same organization,and the structure of the two networks can better understand the formation and development of the disease at the molecular level.In this paper,the gene expression profiles of renal tissues with or without renal clear cell carcinoma were screened and analyzed,and genes with large differences were screened as candidate genes.And then we constructed a mutual information network on the genes obtained by screening.Based on the seven statistics of the network structure,a new concept of "area difference" is proposed,and a new method of screening for structural key genes that may have a significant effect on the disease development process is proposed.Eight Genes as potential pathogenicity genes for renal clear cell carcinoma.Experimental studies of the cancer showed that six of the eight genes were associated with the formation and progression of clear cells in renal cells and that the remaining two genes were expected to have an effect on the development and progression of renal clear cell carcinoma.The method that based on the network structure analysis to study the disease sensitive gene is the subject which of important theoretical significance and practical significance.This approach will be more effective in studying the pathogenesis of cancer and provide a means for deciphering the formation of other non-cancerous diseases
