摘要 目的观察3-磷酸甘油醛脱氢酶(G3PD)修饰蛋白(MP)对肾小管上皮细胞的修复作用,探讨G3PD的MP对庆大霉素所致急性肾功衰竭(acuterenalfailure,ARF)大鼠的保护作用。方法采用庆大霉素制作ARF大鼠模型,随机分成正常对照、阳性对照(简称模型组)及MP治疗组3组。分别于实验第7和14天测定各组血清SCr以及血清、肾皮质匀浆MDA和SOD。结果治疗第7天,MP治疗组SCr水平明显低于模型组(t=4.520,P<0.01),血清及肾皮质匀浆MDA水平明显高于正常对照组(t=10.321,-11.590,P<0.01),但低于模型组(t=3.939,4.609,P<0.01);SOD水平明显降低(t=-18.863~-7.857,P<0.01)。至第14天,上述各项指标均明显恢复与第7天时相比差异有显著意义。MP治疗组的肾组织病理改变程度明显轻于模型组。结论MP具有缓解氧自由基损伤,减少炎性细胞浸润,促进细胞增殖,减轻ARF大鼠肾脏病变及改善肾功能的作用。 Aim To investigate whether MP of G3PD improve the TEC impairing,and t o observe the effect of G3PD modifier protein (MP) in acute renal failure(ARF) r ats with gentamycin.Methods By means of the animal model of ARF induced by genta mycin,SD rats were divided into three groups,i.e.normal control group, model gro up and treatment with MP of G3PD group.At the 7th and 14th days,the SCr of serum ,MDA and SOD of serum and renal tissue were measured and the renal histology was observed by light and electron microscopy.Results After 7 days MP treatment, th e level of SCr of MP group was higher than control group(P< 0.01) but lower comp aring with model group(P< 0.01),MDA in the MP group was lower and SOD was higher than that in the model group(P< 0.01).LM and EM examination showed that renal p athological change of the MP group was slighter than that of the model group.Con clusions MP of G3PD can attenuate of the oxygen free radicals impairing and leuk ocyte infiltration.MP can improve renal pathological change of the ARF rats and ameliorate the renal function.
