赤丹退黄颗粒乙醇提取工艺正交试验研究

摘要 目的 为赤丹退黄颗粒提取工艺提供依据。方法 采用正交设计法 ,因素为溶剂用量、浓度、提取时间和次数 ,以芍药苷及出膏率指标综合分析。结果 最佳组合为A2 B2 C3 D2 ,但考虑实际生产溶剂回收、降低成本、节约时间等因素 ,确定乙醇提取工艺为用 5倍量的 70 %乙醇提取 3次 ,每次 2h ,即A2 B3 C3 D2 。结论 提取时间对芍药苷和出膏率的影响最大 ,溶剂用量、浓度和提取次数三项水平间没有显著性差异 。 Aim To provide a basis for studying the extraction process of traditional medicine Chidan Tuihuang Keli.Methods The orthogonal design with four factors: alcohol amount,alcohol concentration,extraction time and degree were adopted.Paeoniflorin and total extract worked as markers.Results The optimum extracting condition was A 2B 2C 3D 2.However,other factors,such as cost and cycle,were taken into account in actual production.In the end,adding five times amount of 70% alcohol and refluxing and extracting for 6h (totally extracting for 3 times) was confirmed.Conclusion The extraction time influenced paeoniflorin and total extract greatly.The factors alcohol amount,alcohol concentration and extraction degree should not be strictly controlled because they were not insignificant difference in there levels.
