摘要 目的通过观察肿瘤坏死因子-α(TNF-α)和白细胞介素-1(IL-1)作用后大鼠肺微血管内皮细胞(PMVEC)中血小板内皮细胞黏附分子-1(PECAM-1)表达的改变,初步探讨血管内皮黏附分子的功能和TNF-α,IL-1的致炎机制。方法应用免疫组织化学法和流式细胞仪检测TNF-α和IL-1作用后大鼠PMVEC中PECAM-1表达的改变。结果流式细胞仪检测发现,TNF-α作用后大鼠PMVEC中PECAM-1较对照组显著减少,而IL-1作用后大鼠PMVEC中PECAM-1表达无显著改变;免疫组化检测发现,TNF-α和IL-1作用后大鼠PMVEC细胞间连接部的PECAM-1染色较对照组明显变淡。结论TNF-α,IL-1等炎性刺激物可能通过改变内皮细胞中黏附分子的表达和分布而发挥重要作用。 Aim To investigate the function of platelet endothelial cell adhesion molecule 1(PECAM 1) and the mechanism of tumor necrosis factor α(TNF α)and interleukine 1(IL 1) inducing inflammation through observing the effects of TNF αand IL 1 on the expression of PECAM 1 in rat pulmonary microvessel endothelial cell (PMVEC).Methods Flow cytometry and immunohistochemical were used to measure protein expression changes of PECAM 1 induced by TNF αand IL 1 in rat PMVEC.Results The flow cytometry showed that the PECAM 1induced by TNF αreduced more significantly in the experimental group than the control group, and the expression of PECAM 1 induced by IL 1 changed insignificantly.Immunohistochemically, the PECAM 1 staining at the joints of PMVEC induced by TNF αand IL 1 was significantly lighter compared with the control group.Conclusion Inflammatory factors, such as TNF αand IL 1, play an important role in inflammation, causing the protein expression reduction of PECAM 1 in PMVEC,and PECAM 1 may play an important role during inflammation.
