摘要
目的 初步观察培高利特对大鼠黑质多巴胺神经元的保护作用 ,并探讨其可能的机制。方法 通过立体定位注射 6 羟基多巴胺 (6 OHDA)制作帕金森病大鼠模型 ,观察预先注射培高利特对阿朴吗啡诱发的旋转行为及受损侧黑质病理变化的影响。结果 大鼠持续注射培高利特 7d后可明显减轻PD大鼠的旋转行为 ,并对抗 6 OHDA所致黑质多巴胺神经元的减少。而选择性D2 受体拮抗剂氨黄酰基苯甲酰胺化合物 (Sulpiride)可阻断培高利特的保护效应。结论 培高利特对多巴胺神经元具有保护作用 ,其机制可能与刺激多巴胺D2
Objective To investigate whether or not pre treatment with pergolide, could protect rat against 6 hydroxydopamine(6 OHDA) induced injury to dopaminergic(DA) neuron in the substantia nigra.Methods Unilateral PD rat models were made by injecting stereotaxically 6 OHDA.The effects of pre treatment with pergolide on the apomorphine(APO) induced rotational behavior and the pathological changes of substantia nigra on the lesion side were observed.Results Pre treatment with pergolide for 7 days significantly decreased the rotational behavior and the redamage of the DA neurons on the lesion side in PD rat models. Pretreatment with sulpiride,a DA D 2 antagonist,abolished the neuroprotection of pergolide.Conclusion Pergolide could provide neuroprotection to DA neuron, and its mechanism was possibly associated with stimulation of the DA D 2 receptor.
