摘要 研究了采用重结晶法纯化番茄红素过程中 ,溶剂种类、结晶温度和结晶时间对番茄红素纯度和收率的影响 ,并通过扫描电镜、高压液相 -质谱联用色谱仪、红外光谱仪及核磁共振波谱仪对所得番茄红素进行了形貌、结构表征 .结果表明 ,番茄红素为片状晶体 ,以正己烷为重结晶溶剂在 - 5°C下保持 30 min可简洁、高效地获得高纯度番茄红素 .结晶温度高于 - 5°C或结晶时间不足 30 min时 ,收率较低 ;结晶温度低于 - 5°C时 。 The effects of solvent types, recrystallization temperature and recrystallization time on purity and yield of lycopene purified from tomato using recrystallization were investigated. Moreover, the appearance and the molecular structure of lycopene were charaterized. Not only high purity but also high yield of lycopene could be obtained when hexane as solvent, recrystallization temperature (R TEAM) at -5 °C and recrystallization time (R TIM) at 30 min. R TEAM over -5 °C or R TIM under 30 min led to the loss of yield while R TEAM under -5 °C led to the decrease of purity. Additionally, flake shape of lycopene crystals was observed by the scanning electronic microscope (SEM) and the molecular structure of lycopene was characterized by high performance liquid chromatography mass spectrum (HPLC MS), nuclear meganetic resonance (NMR) and infrared spectrum (IR).
