反相高效液相色谱法测定狗血浆中安非他酮的浓度 被引量:3

摘要 目的:建立反相高效液相色谱法测定血浆中安非他酮的浓度。方法:血浆样品经处理后,选用C18柱(150MM×4.6mm,55μm),0.05mol.L-1磷酸二氢钠缓冲溶液(0.1mol·L-1氢氧化钠溶液调pH5.5)-甲醇(44:56)为流动相,紫外检测器(λ=254nm),羟乙基氟西泮为内标。结果:安非他酮血药浓度的线性范围为1.0-500.0ng·mL-1(r=0.9974),提取回收率78.40%-85.70%,方法回收率95.62%-98.82%,日内、日间RSD分别为3.8%-4.9%和2.9%-4.2%。结论:该方法准确、灵敏,适于安非他酮药代动力学研究。
