摘要
DNA疫苗相对于传统疫苗,具有安全、方便、成本低等优点,但大多数MHC分子介导的DNA疫苗免疫途径不能够产生有效的免疫应答,是影响DNA疫苗免疫效果的主要因素之一。利用MHC分子特殊的免疫学功能,将其与抗原表位偶联,形成稳定抗原肽-MHC分子复合体,使"裸DNA"成为专业的抗原呈递分子的目标,并直接呈递给T细胞,能更有效地诱导机体免疫应答,无疑将成为改善DNA疫苗效率的有效途径。介绍了基于MHC分子开发新型DNA疫苗的研究进展,以期为新型核酸疫苗的设计与开发提供新思路。
DNA vaccine is superior to the traditional vaccines in the aspects of safety, convenient usage and low production cost.However, most of the immunization of DNA vaccines mediated by MHC molecules fails to produce an effective immune response,which is one of the main factors limiting the immune effect of DNA vaccine. According to the special immunological function of MHC molecules, stable MHC complexes with antigenic peptides can be constructed. These naked DNA are identified by professional antigen-presenting molecules and directly presented to T cells, which make the immune response more effective.There is no doubt that this will be a promising way to enhance the efficiency of DNA vaccines. The research progress of a new kind of DNA vaccine based on MHC molecule are reviewed in this paper, so as to provide some new ideas for the design and development of nucleic acid vaccines.
