摘要
本研究分离筛选乳酸菌并进行抑菌试验比较研究,采用双层平板法从发面面肥、核桃叶青贮、玉米青贮、棉秸秆黄贮中共分离出31株乳酸菌。通过抑菌实验筛选出发面面肥中的F3和F11,核桃叶青贮中的HTY1. 9,玉米青贮中的YM1. 1和棉秸秆黄贮中的MG1. 5,共5株乳酸菌具有抑菌作用;抑制金黄色葡萄球菌效果最好为HTY1. 9,抑菌直径为15±0. 15 mm,抑制大肠杆效果最明显的是YM1. 1,抑菌直径是11. 5±0. 17 mm;对黄曲霉菌抑菌效果最好的是F11,抑菌直径为25±0. 12mm。经16S r DNA序列分析鉴定,F3与芽孢杆菌的相似性为100%,F11与屎肠球菌的相似性为100%,HTY1. 9与植物乳杆菌的相似性为99. 68%,YM1. 1与布氏杆菌的相似性为99. 52%,MG1. 5与魏斯氏乳酸菌的相似性为99. 32%;具有抑菌作用的5株乳酸菌分别为芽孢杆菌(Bacillus altitudinis)、屎肠球菌(Enterococcus faecium)、植物乳杆菌(Lactobacillus plantarum)、布氏杆菌(Brucella)和魏斯氏乳酸菌(Lactobacillus Weiss)。
In this study,lactic acid bacteria was isolated from Xinjiang traditional sourdough samples,walnut leaf ensiling,corn silage and cotton straw ammonification,and their bacteriostatic tests were carried out. The experiment was used the double deck plate method were isolated from sourdough samples,walnut leaf ensiling,corn silage,cotton straw ammonification,and five of them could inhibit indicative bacteria. The bacteriostatic effect was evaluated by the size of bacteriostasis. The best bacteriostasis of Staphylococcus aureus was HTY1. 9,the diameter of bacteriostasis was 15 ± 0. 15 mm,the most obvious effect of inhibiting the coliform was YM1. 1,the bacteriostasis diameter was 11. 5 ± 0. 17 mm,and The best bacteriostasis effect for Aspergillus flavus was F11 and the diameter of bacteriostasis was 25 ± 0. 12 mm. Based on 16 S r DNA gene sequnce analysis the similarity between strain F3 and Bacillus altitudinis was 100%,The similarity between strain F11 and Enterococcus faecium was 100%,HTY1. 9 and Lactobacillus plantarum had the similarity of 99. 68%,YM1. 1 and Brucella had the similarity of 99. 52%,the similarity between strain MG1. 5 and Weiss lactobacillus was 99. 32%. Five lactic acid bacteria were identified as Bacillus altitudinis,Enterococcus faecium,Lactobacillus plantarum,Brucella and Lactobacillus Weiss.
