摘要
目的 探讨对氧磷酯酶 2 (PON2 )基因 A14 8G多态性与糖尿病视网膜病变的关系。方法 (1)用聚合酶链反应 -限制性片段长度多态性 (PCR- RFL P)分析法探查 PON2基因 A14 8G多态性在正常对照组、单纯 2型糖尿病组、糖尿病视网膜病变组中的基因分布频率 ;(2 )放免法检测血清免疫反应性胰岛素 (IRI)、 C肽 (C- P)水平。结果 (1)糖尿病视网膜病变组的 GG基因型和 G等位基因频率明显高于单纯 2型糖尿病组 (X2 =3.98P<0 .0 5 ,X2 =4 .4 9P<0 .0 5 )和正常对照组 (X2 =8.4 4 P<0 .0 1,X2 =8.6 6 P<0 .0 1) ;(2 )方差分析结果显示基因型为 GG的糖尿病患者空腹血糖浓度明显高于基因型为 GA和 AA的糖尿病患者 (F=3.94 P<0 .0 5 ,F=5 .17P<0 .0 5 ) ,而正常对照组各基因型间空腹血糖浓度无显著性差异 (P>0 .0 5 ) ;(3)非参数检验表明 PON2基因多态性与糖尿病视网膜病变的发生有关 (Z=0 .5 74 P<0 .0 5 ) ,多因素分析结果表明 PON2基因型 (相对危险度为 3.5 4 71P<0 .0 0 1,模型总判对率 90 .31% )和空腹血糖浓度 (相对危险度为 4 .0 14 3P<0 .0 0 1,模型总判对率 89.74 % )与糖尿病视网膜病变独立相关。结论 PON2基因多态性与糖尿病视网膜病变有关 ,糖尿病视网膜病变的易感性在一定程度上与较高的空腹血糖浓度?
Objective To determine whether paraoxonase2(PON2)gene A148G polymorphism is associated with diabetic retinopathy(DR).Methods Genotype distribution and allele frequency of PON2 gene were investigate by means of polymerase chain reaction restriction fragment length polymorphism(PCR RFLP).Serum Immune reactive insulin(IRI)and C peptide(C P)were measured by radio immunity.Results DR patients had a higher GG genotype and G allele frequency than that of simple type 2 diabetes mellitus(T2D)subjects(X 2=3 98 P <0 05,X 2=4 49 P <0 05)and normal control subjects(X 2=8 44 P <0 01,X 2=8 66 P <0 01).In subjects with T2D,the PON2 codon 148 G/G homozygotes had significantly higher mean fasting plasma glucose than subjects with the other two gentypes(F=3 94 P <0 05,F=5 17 P <0 05).However,in normal control subjects,there was no difference in mean plasma glucose among any of the genotypes( P >0 05).In univariate analyses,the paraoxonase 2 polymorphism was associated with DR(Z=0 574 P <0 05).When subjects to multivariate analyses,the paraoxonase2 polymorphism remained statistically associated with DR independent of traditional risk factors( P <0 001).Conclusion The paraoxonase2 gene polymorphism were significantly associated with DR independent of traditional risk factors in T2D patients,Which is associated with higher fasting plasma glucose concentration.
