摘要
目的 探讨联合转染P2 1基因和c -Fos反义核酸对自体移植静脉内膜增殖的影响。 方法 选择 2 0只新西兰家兔 ,随机等分为实验组和对照组 ,均行自体颈外静脉、颈总动脉移植手术 ,仅实验组移植静脉段行腺病毒介导的P2 1基因溶液浸泡、吻合口周围行c-Fos反义核酸凝胶涂布 ;术后 2周取出移植血管 ,分别行病理学、免疫组织化学检查。 结果 实验组移植血管内膜厚度、管腔狭窄度、内膜平滑肌细胞数以及增殖细胞核抗原 (PCNA)阳性表达细胞数均较对照组明显减少。 结论 联合转染P2 1基因反义c -Fos核酸可有效地抑制移植静脉内膜的增生 ,是一种比较有发展前途的防治移植静脉再狭窄的基因疗法。
Objective To investigate the effect of P 21 gene and c-Fos antisence olioneucleotide genetic transfection on the intimal proliferation of autografted veins and search for a better genetic therapy Methods The external jugular veins were autografted into common carotid arteries in 20 rabbits and divided evenly into experiment and control groups.The transplanted veins were immersed in the adenovirus guided P 21 cDNA solution for 15 minutes before anastomosis and coated with c-Fos antisence olioneucleotide glue gel just after anastomosis.The autografted vein were removed and observed pathologically and immunohistochemically determined two weeks later.Results The thickness of the venous intima,the degree of the vascular luminal stricture,the numbers of vascular smooth muscle cells and the expressions of PCNA in the experiment group were less than those in the control group.Conclusion It is suggested that P 21 gene and c-Fos antisence olioneucleotide genetic transfection can inhibit the proliferation of the autografted venous intima.It is a prospective genetic therapy to prevent the restricture of the autografted veins.
