摘要
利用ISSR标记对70份鱼腥草种质资源遗传多样性进行检测。结果发现,所有引物扩增产物均具多态性,22个ISSR引物共得到352条扩增DNA片段,平均每个引物可获得16条DNA片段。其中,92.3%的片段具多态性。每个多态性引物能扩增出4~58条DNA片段,平均可扩增出14.8条多态性片段,峨眉蕺菜与蕺菜中染色体数目为36的细胞型间相似程度最高,其平均遗传相似系数GS值达0.618。蕺菜中,除染色体数目为54的细胞型外,其余细胞型的类间GS值几乎均是与染色体数目为36的细胞型间最小。栽培蕺菜类群遗传多样性略高于野生类群。聚类分析表明,利用ISSR技术可将全部供试材料区分开,所有材料共划分为8类。根据ISSR遗传相似系数划分的类群主要同染色体数目有关,也与地理分布有一定关系。据此认为,鱼腥草种质资源在分子水平上确实存在较大遗传差异,ISSR标记可作为构建鱼腥草DNA指纹图谱的有效工具之一。鱼腥草药材道地性与环境因素有关,但更大程度上由其遗传因素所决定。本文中还讨论了蕺菜属植物的起源演化。
The genetic diversity of To germplasm resources of Cordate Houttuynia has been tested by ISSR marker and it is found that all the cloning matters of primers have their polymorphism,352DNA cloning fragments from22ISSR primers,of which each produces16DNA fragments in average and92.3%of them are given polymorphism.Each of the polymorphous primers is able to clone4~58DNA fragments,averaging14.8.For Houttuynia emeiensis and Houttuynia cordate those who have the same chromosome number of36are most similar in their cellular types and the value of their genetic semilarity factors(GS)average0.618.The GS value of almost all the cellular types of Houttuynia cordate are smaller than those with their Chromosome number of36excepting the cellular types with chromosome number of54.The genetic diversity of cultivated species of Houttuynia cordate is slightly higher than that of natural ones.Cluster analysis shows that all the materials of8sorts for test can be discrimimated by ISSR technology.The species divided according to ISSR genetic similarity factors are mainly associated with chromosome number and to some extent with their geographic distribution.Therefore it can be consisdered that great genetic defference certainly exists at the cellular level in the germplasm resources of cordate Houttuynia and ISSR marker can be taken as anefficient tool for the establishment of its DNA fingerprint.As a Chinese medicinal plant cordate Houttuynia has its genuineness related to its environmental factors but to a greater extent determined by its genetic factors.In this article the origination and evolution of genera Houttuynia cordate is discussed as well.
