异紫堇碱对人子宫内膜腺癌细胞系-1B细胞增殖、凋亡及迁移的影响 被引量:5

摘要 目的探索异紫堇碱对人子宫内膜腺癌细胞系-1B (HEC-1B)细胞增殖、凋亡及其迁移的影响。方法噻唑蓝比色法检测细胞增殖抑制情况;Annexin V-FITC/PI凋亡检测试剂盒检测细胞凋亡情况;Transwell小室迁移实验检测细胞迁移能力的变化;细胞免疫荧光检测细胞核因子κB (NF-κB)进核情况;Western blotting检测细胞中Bax、Bcl-2、MMP2、MMP9蛋白及细胞核内NF-κB(p65)蛋白的变化。结果异紫堇碱可时间-浓度依赖地抑制HEC-1B细胞的增殖,并浓度依赖地抑制细胞迁移,诱导细胞凋亡。进一步在分子水平上,异紫堇碱可降低HEC-1B细胞中Bcl-2、MMP2、MMP9蛋白的表达,抑制NF-κB进核,增加Bax的表达。结论异紫堇碱可抑制HEC-1B细胞增殖、迁移,诱导细胞凋亡,这种抑制与诱导作用可能与异紫堇碱抑制NF-κB激活进核相关。 Objective To explore the effects of isocorydine on proliferation,apoptosis and migration of human endometrial adenocarcinoma cell-1B(HEC-1B).Methods MTT assay was used to detect cell proliferation.Annexin V-FITC/PI apoptosis assay kit was used to detect apoptosis.Transwell chamber migration assay was used to detect cell migration ability and cell immunofluorescence was used to detect NF-κB in cell nucleus.The changes of Bax,Bcl-2,MMP2,MMP9 protein and NF-κB(p65)protein in the nucleus were detected by Western blotting.Results Isocorydine inhibited the proliferation of endometrial cancer cell line HEC-1B in a time-concentration dependent manner and inhibited cell migration in a concentration-dependent manner,thus inducing apoptosis.Furthermore,at the molecular level,isocorydine could decrease the expression of Bcl-2,MMP2 and MMP9 proteins in HEC-1B cells,inhibit the nucleation of NF-κB,increase the expression of Bax.Conclusion Isocorydine can inhibit the proliferation and migration of HEC-1B cells and induce apoptosis.This inhibition and induction effects may be related to the inhibition of NF-κB activation by isocorydine.
