摘要
目的 探讨舌鳞状细胞癌Tca - 8113细胞纤维连接蛋白受体α5β1的表达 ,及α5β1在Tca - 8113细胞粘附、运动中的作用。方法 培养Tca- 8113细胞 ,采用间接免疫荧光检测细胞α5β1的表达 ;应用抗α5β1抗体结合细胞表面纤维连接蛋白受体α5β1,分析α5β1在细胞粘附、运动行为中的作用。结果 α5β1在舌鳞状细胞癌Tca -8113细胞表面呈点状和团块状分布 ;抗α5β1抗体可以显著抑制细胞在纤维连接包被表面的粘附和运动行为。结论 舌鳞状细胞癌细胞存在α5β1的表达 ,α5β1在舌鳞状细胞癌的浸润、转移中可能起着重要的作用。
ObjectiveTo investigate the expression of α 5β 1 in Tca-8113 cell line of tongue squamous cell carcinoma and its roles in adhension and migration of the cells.MethodsTca-8113 cells were cultured with RPMI-1640 medium in vitro.Expression of α 5β 1 in Tca-8113 cells was detected by indirect immunoinfluorescence.Incubated with rabbit anti-human α 5β 1 antibody,the adhesion and migration of the cells on fibronectin coated surface were analyzed.ResultsTca-8113 cells could produce α 5β 1 and showed brightly dot or punctate staining over entire cellular surface when examined by immunofluorescence with antibody to α 5β 1.Antibody to α 5β 1 could significantly decrease the adhesion and migration of Tca-8113 cells on fibronectin coated surface.ConclusionTca-8113 cells can synthesize α 5β 1 on the cellular surfaces,and α 5β 1 may play an important role in the invasion and metastasis of tongue squamous cell carcinoma.
