摘要
目的 了解异丙酚对大鼠肝脏一氧化氮合酶的影响。方法 40只SD大鼠 ,随机分为异丙酚组、对照组 ,分别腹腔注射等容积异丙酚 (10ml/kg ,即 10 0mg/kg)和生理盐水 (10ml/kg)。异丙酚组待鼠翻正反射消失后 ,微泵输注异丙酚 ,2 0min后处死 ;对照组鼠腹腔注射 2 0min后处死。检测肝脏组织匀浆中NO水平、NOS酶活性及内皮型NOS(eNOS)在肝脏内的表达与分布 (免疫组化法 )。结果 异丙酚组肝脏组织匀浆中NO水平、NOS酶活性均明显高于对照组 (P <0 0 1)。免疫组化结果显示 :异丙酚组肝脏血管内皮细胞eNOS染色表达强阳性 ,半定量比较有显著差异 (P <0 0 1)。结论 异丙酚可以刺激肝脏中NOS活性 。
Objective\ To investigate effects of propofol on nitric oxide synthase in rat liver.Methods\ 40 SD rats were randomly assigned to propofol group(n=20) given enterocoelia injection propofol(10ml/kg,100 mg/kg) and control group(n=20) given enterocoelia injection saline(10 ml/kg). The rats in propofol group infused propofol by micropump after the disappearance of reflex action were killed 20 mins later,and the rats in control group were killed 20 mins after enterocoelia injection. Then,the levels of NO and the activity of NO synthase (NOS) in liver constitution homogenate were measured respectively. At the same time,the cellular distribution and expression of endothelial NOS(eNOS) in liver were investigated by using immunohistochemical technique .Results\ The levels of NO and the activity of NOS in liver constitution homogenate of propofol group were higher than that of control group(P<0.01). The results of immunohistochemistry indicate that the expression of eNOS in propofol group was more obviously positive than that of control group (P<0.01). Conclusion\ Propofol can stimulate the activity of NOS in liver and raise the level of endogenous NO.
