摘要
目的 比较红霉素治疗前后绿脓杆菌菌落中藻酸盐含量的变化 ,了解红霉素对绿脓杆菌生物被膜的作用。方法 利用藻酸盐经强酸处理后生成的糠醛或其衍生物 ,再与酚类、胺类化合物作用生成有色物质的原理 ,采用比色法测定红霉素治疗前后绿脓杆菌菌落中的藻酸盐含量 ,并测定比色法的准确性和重复性。结果 菌落中的藻酸盐含量与反应物的吸光度值 (A)呈正相关 (r =0 .98,P <0 .0 1) ;比色法的回收率为 98.18% ,变异系数小于 5 % ;红霉素治疗前后绿脓杆菌菌落的藻酸盐含量分别为 2 3.5 4和 18.0 2 μg/ml,差异有显著性 (P <0 .0 5 )。结论 比色法的准确性和重复性均较高 ;
Objective To detect the alteration of the content of alginate colony of pseudomonas aeruginosa after the treatment by erythromycin, so as to learn the effect of erythromycin on biofilm formed by pseudomonas aeruginosa . Methods Determine the content of alginate in colony of pseudomonas aeruginosa by colorimetric method and analyze the precision and reproducibility of the method. Results The content of alginate was positively correlated with the optical density of the mixture( r =0.98, P < 0.01 ). The rate of recovery was 98.18%, and the coefficient of variation was less than 5%. The contents of alginate before and after treatment were 23.54 and 18.02 μg/ml.Conclusion This method gives good precision and reproducibility. The content of alginate decreased after treatment with erythromycin.
