摘要
目的:探讨NGAL在腺嘌呤致大鼠肾纤维化中的表达情况。方法:将48只Wistar雄性大鼠,随机分为2组。即正常组(n=24)、模型组(n=24),模型组每天予以200mg/kg的腺嘌呤灌胃,正常组摄入等量的NS,连续灌胃6周,在第2、4、6周末将肾组织取出,通过HE染色观察病理改变,免疫组化染色观察大鼠肾组织中NGAL的表达情况,应用图像分析系统软件进行半定量分析。结果:正常组大鼠肾组织结构正常,模型组在第2周时表现管腔扩张,肾小管出现肿胀,肾间质有弥漫性炎性细胞浸润,间质出现纤维化,第4周时肾小管扩张更明显,肾间质仍有较多的炎性细胞浸润,肾间质纤维化较前明显,第6周时肾小管萎缩甚至有坏死,肾间质广泛纤维化。NGAL在正常对照组大鼠肾组织有少量的表达,尤其以肾小管表达为主,与正常组比较,模型组第2周时肾组织中NGAL的表达水平显著升高,有统计学意义(P<0.05),第4、6周时NGAL的表达水平明显低于第2周。结论:NGAL在腺嘌呤所致大鼠肾纤维化的表达水平显著增高,与对照组比较差异显著,提示其可作为肾纤维化发展的指标,为临床上判断病情提供参考。
Objective: To evaluate the expression of NGAL in kidney tissue of rats with renal fibrosis. Methods: 48 male Wistar rats were randomly divided into 2 groups: the normal group( n = 24) and model group( n =24). Rats in model group were given 200 mg/kg adenine everyday,while normal group were given the same amount of NS. The renal tissue was removed on the 2th,4th,6th week. Then we observed the pathological changes by HE staining,and evaluated the the expression of NGAL by making a semi- quantitative analysis with an image analysis system software. Result: normal rat renal tissue structure was normal,compared with the normal group. Model group showed luminal dilatation at the second week,the renal tubular swelling,renal interstitial diffuse infiltration of inflammatory cells,interstitial fibrosis. The tubular dilatation was more obvious at the fourth week,renal interstitial still intensive inflammatory cell infiltration,renal interstitial fibrosis significantly. At the sixth week of tubular atrophy and necrosis,renal interstitial extensive fibrosis. NGAL in normal renal tissue of rats had a small amount of expression,particularly in the renal tubules,compared with the normal group. The expression levels of model group at the 2th week were significantly higher( P < 0. 05). At the 4th and 6th week,NGAL expression levels were significantly lower than those at tthe 2th week. Conclusion: NGAL expression levels in adenine- induced renal fibrosis in rats was significantly higher,and it may suggests that it can be used as indicators for the development of renal fibrosis,and it can provide a reference for judging the clinical condition.
