摘要
目的:探讨一种壳聚糖-胶原-BMP2微球复合材料在制备时的规律特点,为今后组织工程中生物材料的制备提供理论基础。方法:水包水法制备BMP-2微球;冷冻干燥法制备不同比例组合的BMP-2微球-壳聚糖/胶原复合支架;乙醇置换法检测复合支架孔隙率;平衡溶胀试验测量吸水率;细胞培养后测量相关收缩率;取壳聚糖和胶原4/6、1/9比例的样本做扫描电镜观察和MTT实验。结果:各比例材料的孔隙率稳定;一定范围内吸水率随胶原比例增加而增加;收缩率随壳聚糖比例增加而减小;壳聚糖和胶原4/6时孔径均匀,促细胞增殖能力较强。结论:BMP-2微球复合壳聚糖-胶原支架中壳聚糖/胶原以比例4/6相复合时性能较佳。
Objective:To invesitigate the characteristics of preparation of BMP-2 loaded chitosan/collagen scaffolds for tissue engineering.Methods:The microspheres were prepared by w/O/w;The different proportions of scaffolds were prepared by lyophilization;The porosity,swelling rate and CMC were determined by some tests and formulas;The scaffolds of ratios of 4/6 and 1/9 were selected to be observed by SEM and MTT experiment.Results:There is no difference at porosity among the experiment groups.The swelling rate increased with increased collagen proportion.The CMC decreased with increased chitosan proportion.The scaffold at ratio of 4/6 shows more homogeneous and better ability to promote cell proliferation.Conclusions:The BMP-2 microspheres-chitosan/collagen scaffold prepared at proportions of 4/6 shows better characteristics.
